چکیده

Green tiger prawn, Penaeus semisulcatus, is the most abundant and important prawn in the Persian Gulf. This species belongs to Penaeus genus and Penaeidae family. Based on body and wipe antenna color pattern, two distinct populations of this species have been reported in the Persian Gulf. Dept morphological and meristic studies along with the electrophoresis pattern of proteins from various tissues, have led to identification of these two populations as the new subspecies, Penaeus semisulcatus persicus. In the past, mitochondrial genome has been used as an important marker to determine the phylogenic relations among similar species. Therefore, in this study, a fragment of mitochondrial genome (4050bp) was amplified to evaluate the phylogenic relationship among of green tiger prawn and its subspecies. To amplify the above fragments primer pairs mtF6/mtR6, mtF7/mtR7, mtF8/mtR8 were used. The aforementioned primers led to the amplification of fragments of the size 1800, 1790, 1470, respectively. Upon determining the nucleotide sequence, it was specified that partial sequence of mitochondrial genome of the sizes 4056 and 4049 were in species and subspecies, respectively. The above section include the NAD4, NAD4L, tRNA-Thr, tRNA-Pro, NAD6, Cytb and tRNA-Ser genes and partial sequence of NAD1 gene. The phylogenetic relationship between the species and subspecies and also other similar species was studied using the NAD4, NAD6, CytB genes and partial sequence NAD1 gene. The genetic distance between the species and subspecies were calculated to be 0.136, 0.191, 0.168, 0.185, respectively, based on the above genes. The genetic distance was obtained to be 0.167±0.007 based on complete nucleotide sequence. Genetic distance calculated the genes among a number of Penaeus genus was less than the genetic species and subspecies.
