چکیده

The research was carried out as factorial experiment in a completely randomized design. In this research, the callus induction of two varietys of hybrid cucumber seeds and the comparison of their morphological and biochemical responses to independent stress of sallinity in in vitro culture were investigated. At first, seeds of two varietys were prepared and transferred to the laboratory in MS medium culture medium. Cotyledon and hypocotyle explants for calluse induction to MS-containing glasses were combined with different treatments of growth regulators of auxins and cytokinin such as BA at concentrations (0, 0.5 and mg/l) and 2,4-D and NAA were transferred with concentrations (0, 0.5, 1 and 2 mg/l ) with three replications and then in a growth chamber at 25± 3°C to observe changes Morphological (colour, texture and relative growth of calli) were kept. Moreover, among different combination and concentrations of PGRs tested, MS 1 mg/l NAA and 0/5 mg/l BA showed the highest rate of callus regeneration. The highest rate of callus regeneration was obtained from hypocotyle . The obtained callus from both explants were green and compact. In order to proliferation, the apical bud explants budding was combined with the combination of growth regulators BA (1, 2 and 3 mg/l) and IAA (0 and 0.5 mg/l). The results showed that the highest number of shoots was with the use of one milligram per liter of BA. For root production. Rooting treatments NAA with concentration of (0, 0/5, 1 and 1/5 mg/l ) were tested. Results showed that the was no significant difference between the two varietys for root number and length.Hypocotyl explants were used to produce embryonic callus in MS medium containing 0.5 mg/l BA and 1 mg/l NAA After one month, calluses were cultured in MS medium containing 2 mg/l BA and 0.3 mg/l NAA after about 4 weeks of seedling production.The best calli were selected for in vitro salinity (0, 20, 40 and 60 mM sodium chloride). After one month, some biochemical charact
