Initial laboratory studies (In vitro) investigations and determination of optimal percent of combined probiotics and prebiotics before their use in field conditions on basis effective criteria to growth bacteria is a new step for the selection of optimal applications in aquaculture. The present study was conducted to evaluate the effect of different percent of Mananoligosaccharide and Fructooligosaccharide on growth of Pediococcus acidilactici.Probiotc inoculated in a minimal MRS (mMRS) broth medium was modified by omission of glucose supplemented with different levels(0/5%,1% and 2%) of prebiotic carbohydrates. All modified media were adjusted to pH 6/9.Then a quantity of 107and1010 cfu/ml of the above probiotics were inoculated in condition aerobia and anaerobic. During 24 hours of incubation at 37 °C,optical density in interval 2h and pH were determined using spectrophotometer and pH meter. Sodium thioglycollate (0.2%, w/v) was used to create anaerobic condition. The results indicated that mananoligosaccharide were no significant influence on growth P.acidilactici (p>0.05) whereas Fuctooligosaccharide there were significant influence in aerobic and anaerobic condition(p<0.05). The best in vitro growth under aerobic and anaerobic culture condition was observed in the treatment with 0.5% Fructooligisaccharid and 107cfu/ml P.acidilactici.In this study indicated that levels and carbohydrate sources can be effective on growth P.acidilacrici
