Abstract

Background: Very limited research has been done on the isolation and identification of exosomes from marine organisms, and according to the many studies conducted, there has been no report of their extraction from macroalgae. This research is a step to start extracting them from macroalgae. The conducted research has determined that exosomes play an important role in intercellular communication as well as the spread of diseases, so more studies in this field seem necessary to fight diseases. Considering the many uses of exosomes in various diagnostic and therapeutic fields and their many uses in the production of vaccines and new drugs for incurable diseases, it seems necessary to find methods for their economic and mass production that isolate and identify them. It will help to reach this goal in the growth of native algae of the Persian Gulf. Aim: extracting and isolating exosomes from macroalgae and determining some of their characteristics Methodology: The brown alga pavonica Padina was collected from the coastal area of Bushehr, and the exosomes were extracted and purified using an exosome isolation kit. Transmission electron microscope, scanning electron microscope, and dynamic optical measurements were used to determine the morphological characteristics of exosomes. Extracted was done. Cytotoxicity test, flow cytometry, cell migration test, antioxidant and antibacterial test and western blotting were done to check their properties and characteristics. Findings: The exosomes extracted and purified using the kit showed a spherical morphology with an approximate diameter of 100-30 nm in imaging with scanning and transmission electron microscopes, and the size of the particles by dynamic optical measurement was approximately 1-150 nm in size distribution. Showed . Western blot analysis confirmed the expression of two specific exosome markers, CD9 and HSP70, which are expressed on the surface of exosomes at 24 and 70 kDa in exosomes. Exosomes completely inhibited
