Abstract

Introduction & Objective: Enoxacin and ciprofloxacin are two antibiotics with a wide range of effects. Also, by influencing the processing pathway of micro RNAs (miRNAs), they have shown anticancer activity against various types of cancer cells. However, the effect of these two antibiotics on bladder cancer cells has not been studied. In this study, we investigated the effect of these two antibiotics on the 5637 cell line separately and in combination with the anticancer drug cisplatin. Materials and Methods: The effect of enoxacin (124 μM) and ciprofloxacin (150 μM) on viability, cell migration, colonization and apoptosis of 5637 cell line was evaluated. The expression change of some miRNAs in treated and control cells was measured by real time PCR. Results: The results of this research showed that enoxacin at a concentration of 124 μM and ciprofloxacin at a concentration of 150 μM lead to a decrease in viability, colony-forming power and cell migration in 5637 cell line. It was also observed that enoxacin, ciprofloxacin and cisplatin induce cell death in about 80.3, 71.86 and 96.9% of cells in 72 hours, respectively. The combined use of fluoroquinolone antibiotics with cisplatin also had a synergistic effect on reducing the growth and proliferation of 5637 cells. So that the growth of cells treated with the combination of cisplatin and enoxacin decreased by 69% and with the combination of ciprofloxacin and cisplatin by 64%. The expression of has-mir-34a and has-mir-145 miRNAs also increased under the influence of enoxacin. Discussion & Conclusion: According to the obtained results, Enoxacin can be considered as a new drug candidate for bladder cancer.
