Micro-grafting, isone of theadvancedtechniquesin the production ofgrafted treesofdisease. In this research, for preparation of ro0tstock the effects of medium supporting systems including paper bridges, perlite, vermiculite with half concentration of MS and paper bridge with full concentration and with 60 and 75 g sucrose per liter for seed germination of four species of citrus, including Orange Shiraz, large orange, mexican lime and rough lemon was examined. The results of the interaction between the type of seed, of support systems medium culture and concentration of sucrose on seed germination show The highest germination related to mexican lime in perlite medium culture and paper bridge with half MS and with a concentration of 60 g/l sucrose was obtained. The lowest rate germination for all seeds related to medium culture paper bridgewith complete MS and 75 g/l sucrose was observed. In order to provid scion type and Rate of plant growth regulators onlime proliferation and callus induction vere check out. Therefore, young shoot tip culture after the sterilization were placed in culture media prepared. Culture medium containing growth regulators BA with concentrations of 5.0, 1 and 2 mg per liter and NAA with concentrations of 0, 5.0 and 1 mg per liter were prepared. after 5 weeks, results showed the highest number of buds proliferation of buds cultivated mexican lime in a medium containing 1 mg per liter BA, without NAA was obtained.
