Abstract St Johns Wort (Hypericum perforatum L.) from Hypericaceae family is an important medicinal plant, which its secondary metabolites, hypericin and hyperforine, have several medicinal effects such as antidepressant, antiviral, antibacterial and etc. In this study callus induction of two explants of St. Johns Wort including leaves and shoots were evaluated at different level of growth regulators (zero, 2 and 4 mg/l BA, and 0.1 and 2 mg/l kinetin plus 0.1, 0.2 and 0.3 mg/l 2,4-D) in MS. To evaluate shooting zero, 0.5 and 1 mg/l BA and zero, 0.5 and 1 mg/l kinetin were used. In order to produce root, growth regulator IBA at concentrations of zero, 0.2, 0.4, 0.6, 0.8 and 1 mg/l were used. After 4 weeks at 21°C and 16 hours light, the callus induction, shooting and rooting were measured. Data analysis indicated that the highest patch level of 5% in the number of leaf explants achieved at 0.3 mg/l of 2,4-D. Highest mass of callus observed at single node explants in media containing 2 mg/l BA with 0.1 mg/l of 2,4-D, respectively.
