Abstract Yellow fin sea bream is a widespread fish which is important as a commercial fisheries and aquaculture species in south east Asia and Persian Gulf region. In this study Mitochondrial DNA at the cytochrome oxidase 1 (Co-1) gene region was sequenced to molecular identification and comparison with Acanthopagrus latus specimens from other regions. Partial sequencing of COI gene was performed by polymerase chain reactions using general primers. Bioedite ,MEGA and DNasp sofwares were used to analyze sequenced nucleotides. The results of the present study confirms the reported belonging of studied fish to Acanthopagrus latus species . Specimens of the study were in close relationship to Indian Ocean yellow fin sea breams, whereas Japan and China specimens were calculated to be in different cluster of depicted phylogenetic trees. It seems that Indian Ocean and Persian Gulf yellow sea breams are more ancestral than their Japanese and Chinese relatives. DNA barcoding of other sparids of Persian Gulf is suggested using COI sequences.
