Abstract

Background and Purpose: The purification of DNA is one of the important matters in known biology research because DNA purification provides the basis for conducting downstream experiments. One of the new methods of DNA purification can be mentioned using magnetic nanoparticles. The purpose of this research is the synthesis of cobalt ferrite magnetic nanoparticles coated with polyethylene glycol (PEG) and polyethylene imine (PEI) in the purification of free, and genomic DNA from adenocarcinoma gastric cell line (AGS) and comparing it with a column kit. Methodology: First, the AGS cell line was cultured in the cell culture laboratory, cobalt ferrite magnetic nanoparticle was synthesized by hydrothermal method from iron and cobalt chlorites and coated with PEG and PEI polymers, then by X-ray diffraction spectroscopy, infrared conversion spectroscopy Fourier, vibrating sample magnetometer, and scanning electron microscope examination determined the characteristics. After optimizing the concentration of cobalt ferrite magnetic nanoparticles, the appropriate concentration of nanoparticles for DNA extraction is determined, the DNA extraction process with three repeats for each nanoparticle and a column kit from the culture medium (for free DNA extraction) and AGS cell line (for genomic DNA extraction) Done. Recently, polymerase chain reaction (PCR) using actin gene primers and agarose gel electrophoresis were used to check the quality of DNA extracted by nanoparticles and column kit. Results: The results obtained from X-ray diffraction spectroscopy, and Fourier transform infrared spectroscopy show the formation of cobalt ferrite nanoparticle structure and its coating by PEG and PEI polymers. The scanning electron microscope images show the formation of the spherical structure of nanoparticles, the size distribution of nanoparticles shows that the average diameter of cobalt ferrite nanoparticle is 42.35 nm, cobalt ferrite nanoparticle with PEG coating is 55.15 nm, and cobal
