Background: The application of naturally-occurring substances for the treatment of diseases is increasing due to less side effects. Sea cucumbers have long been used to treat diseases, especially the treatment of impotence. Aim: Protective Effects of Persian Gulf Sea Cucumber Holothuria Leucospilota Saponin on Infertility in Male Rats.
Methodology: Sea cucumber saponins Holotoria leucospilota were extracted according to the Hu method with some modifications. Salkovsky tests and infrared spectroscopy(FTIR) were used to identify the extract. In order to investigate the effect of saponins on fertility, 40 Wistar albino rats were divided into 4 groups. Infertility was induced by intraperitoneally administration of 20 mg / kg lead acetate for 4 weeks. For 4 weeks, the treatment was performed in two groups with doses of 10 and 15 mg/kg of extracted saponins. One control group was treated without infertility and one group with lead acetate and no saponin treatment. In the middle and at the end of 8 weeks, qualitative and quantitative tissue tests of the testicles and histochemical blood were performed. The raw results were analyzed using SPSS software and a significant level of 5%.
Result: At the end of the 6th and 8th weeks in the testicular tissue analysis section, with both doses, the diameter of the seminiferous tube, the thickness of the germinal epithelium and the ratio of the area of interstitial tissue to the total tissue, as well as the percentage of abnormal sperm morphology in the head and tail abnormalities sperm motility status in the case of fast and immobile sperm motility, both groups treated with saponin (10,15 mg/Kg) showed a significant improvement compared to the control group(P ? 0.05). On the other hand, studies on the number of leukocytes and the amount of serum CRP protein showed that a significant improvement in parameters in both groups treated with saponins compared with the control group was observed only at the end of the 8th week in the number
